Description Recombinant full-length human Yes, containing additional N-terminal His6 tag. Expressed by baculovirus in Sf21 cells, and purified using Ni2+/NTA-agarose.Yes is a member of the Src family of non-receptor tyrosine kinases and amplifies signals generated by various receptors. Expression of Yes is elevated in melanocytes and in melanoma cells. Yes activity is stimulated by neurotrophins, which are mitogenic and metastatic factors for melanoma cells. Yes is essential for MAP Kinase activation by angiotensin II.Specific Activity: ~1425U/mg where one unit equals the incorporation of 1nmol of phosphate per minute into 0.1mg/ml of Poly (Glu4-Tyr) substrate, at 30?C with a final ATP concentration of 100uM.Applications:Yes Kinase Assay: 1-10ng of this lot of enzyme phosphorylated 0.1mg/ml of Poly (Glu4-Tyr) substrate in the assay described on page two. Assay background was 89,736 CPM, and was subtracted from the actual counts to yield the results.Storage and Stability:May be stored at 4?C for short-term only. For long-term storage, store at -20?C. Aliquots are stable for at least 12 months at -20?C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
