Purity: Highly Purified. C-RESQSYLVEDLERS, C TerminusControl peptide corresponding to polyclonal antibody, Catalog#A3598-05B, goat x human.Alternate Name: PYCARD, PYD and CARD domain containing, ASC, CARD5, MGC10332, TMS1, apoptosis-associated speck-like protein containing a CARD, caspase recruitment domain protein 5, target of methylation-induced silencing-1Apoptosis is regulated by death domain (DD) and/or caspase recruitment domain (CARD) containing molecules and a caspase family of proteases. CARD containing cell death regulators include RAIDD, RICK, BCL10, Apaf-1, ARC, caspase-9, and caspase-2. A novel CARD domain containing protein was recently identified in human and mouse and designated ASC and TMS1 (1R3). Ectopic expression of ASC/TMS1 induced apoptosis through activation of caspase-9 and inhibited the survival of human breast cancer cells (3,4). Overexpression of ASC/TMS1 induced DNA fragmentation (4). ASC/TMS1 is expressed in a variety of human and mouse tissues (1, 2)Application(s):Suitable for use in Peptide ELISA. Other applications have not been tested. Recommended Dilution:Optimal dilutions to be determined by the researcher.Storage and Stability:May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.Peptide Blocking (see corresponding antibody A3598-05B):Antibodies are typically supplied at 0.5mg/ml and peptides as a 100ul pellet. When peptides are reconstituted in 200ul water, the concentration would be also 0.5mg/ml. To start, the best ratio would be 1:1 (which means molar excess of peptides relative to antibodies when identical volumes are mixed). Mix equal volumes of peptide and antibody at the required dilution and leave at ambient temperature. It is best is to have two identical blots, to be incubated with equal amount of antibodies, but one with the antibodies pre-adsorbed to the peptide for 20min. Then incubate and develop the two blots in parallel. |
