| Description | To analyse the I/D- polymorphism, the variable region of the gene is amplified using realtime PCR (e.g. the LightCycler system). Genomic DNA serves as a template for thisamplification. Using this amplicon a melting curve analysis is performed that incorporatesspecific FRET-hybridisation probes detected at 610 nm. This melting curve analysisfacilitates the distinct identification of the I/D genotype. For the real time PCR reaction aprimer is used that flanks the variable region in the gene and produces a fragment. Inaddition two probes (fluorescent labeled sequence specific oligonucleotides; anchor andreporter probe) are incorporated into the PCR reaction mix. Both bind the target DNA inclose proximity in a region between the primer pair. Upon this binding a fluorescent signalis generated that is detected by the optical unit of the real time PCR instrument (e.g. theLightCycler). Note: Do to the test design, there is no increase in fluorescence during thePCR step for samples with a homozygous Insertion. |
