| Description | To analyse the G20210A polymorphism, the variable 3' UTR region of the prothrombingene is amplified using real time PCR (e.g. the LightCycler® system). Genomic DNAserves as a template for this amplification. Using this amplicon a melting curve analysis isperformed that incorporates specific FRET-hybridisation probes. This melting curveanalysis facilitates the distinct identification of the 20210G or 20210A genotype. For thereal time PCR reaction a primer is used that flanks the variable region in the prothrombingene and produces a fragment. In addition two probes (fluorescent labeled sequencespecific oligonucleotides; anchor and reporter probe) are incorporated into the PCRreaction mix. Both bind the target DNA in close proximity in a region between the primerpair. Upon this binding a fluorescent signal is generated that is detected by the optical unitof the real time PCR instrument (e.g. the LightCycler®) at 640 nm.4 2014 Immundiagnostik AG / Version 1.2 / 02.09.2014For the genotyping analysis a melting curve is recorded after the amplification reaction.This is achieved by constantly increasing the temperature. The reporter probe containedin this kit is a perfect match for the wildtype allele and hence requires a highertemperature to be melted from wildtype DNA than from mutated DNA, as here amismatch occurs. The sequence of the reporter probe matches the non-mutated DNA(20210G) perfectly. The hybrids of probe and target DNA will melt at lower temperatures ifthe DNA contains the 20210A variant. In case of a heterozygous sample (only one of thetwo alleles carries the 20210A mutation), two peaks will be generated, one at the lowertemperature (melting point of the mutation) and one at the higher temperature (meltingpoint of the perfectly matched wildtype). |
