| Description | This ELISA is designed for the quantitative determination of S100A12. A monoclonal antibody specific for S100A12 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and the immobilised antibody binds any S100A12 present. After washing away any unbound substances, an HRP-conjugated polyclonal antibody specific for S100A12 is added to the wells. Following a wash to remove any unbound antibody HRP-conjugate, the remaining conjugate is allowed to react with the HRP substrate tetramethylbenzidine. The reaction is stopped by addition of acidic solution and absorbance of the resulting yellow product is measured at 450 nm. The absorbance is proportional to the concentration of S100A12. A dose response curve of the absorbance unit (optical density, OD at 450 nm) vs. concentration is generated, using the values obtained from the standard. S100A12, present in the patient samples, is determined directly from this curve. Range: 0.66 - 54 ng/ml |
