| Description | This enzyme immunoassay is a sandwich assay for the quantitative determination ofanti-tissue transglutaminase antibodies (IgA anti-htTG). The wells of the microtiterplate are coated with the antigen. In a first incubation step, the anti-htTG IgA antibodiesare bound to the coated antigen. To remove all unbound substances, a washingstep is carried out.In a further incubation step, a peroxidase-labeled antibody is added. After anotherwashing step to remove all unbound substances, the solid phase is incubated withthe peroxidase substrate, tetramethylbenzidine (TMB). Finally, an acidic stop solutionis added to terminate the enzymatic reaction, whereby the colour changes fromblue to yellow. The intensity of the yellow colour is directly proportional to the IgAanti-tissue-transglutaminase antibody concentration. A dose response curve of theabsorbance unit (optical density, OD at 450 nm) vs. concentration is generated usingthe values obtained from the standards. IgA anti-tissue transglutaminase antibodiespresent in the samples are determined directly from this curve. |
