Description | To analyze the polymorphism, the variable region of the UGT1A1 gene is amplified usingreal time PCR (e.g. the LightCycler® system). Genomic DNA serves as a template forthis amplification. Using this amplicon a melting curve analysis is performed thatincorporates specific FRET-hybridisation probes. This melting curve analysis facilitatesthe distinct identification of the UGT1A1*28 allele.For the real time PCR reaction in the LightCycler® capillary a primer pair is used thatflanks the variable region in the UGT1A1 gene and produces a fragment with a length of181 bp. In addition two probes (fluorescent labeled sequence specific oligonucleotides;anchor and reporter probe) are incorporated into the PCR reaction mix. Both bind thetarget DNA including the polymorphism in close proximity in a region between the primerpair. Upon this binding a fluorescent signal is generated that is detected by the optical unitof the real time PCR instrument (e.g. the LightCycler®).For the genotyping analysis a melting curve is recorded after the amplification reaction.This is achieved by constantly increasing the temperature. Perfect homologous probesrequire a higher temperature to be melted as those having a mismatch (the sequence ofthe reporter probe and target DNA matches to 100%). The sequence of the reporter probematches the mutated DNA (UGT1A1) perfectly. Hence the melting curve of the TA6-variation (wild type) occurs earlier needing a lower temperature to achieve the maximumas the mutation allele. In case of a heterozygous sample (only one of the two allelescarries the mutation), two peaks will be generated, one at the lower temperature (meltingpoint of the wild type) and one at the higher temperature (melting point of the perfectlymatched mutation). |
