| Description | This ELISA is designed for the quantitative determination of klotho. The assay utilisesthe sandwich technique with two selected monoclonal antibodies that bind to humanklotho.Assay standards and pre-diluted samples which are assayed for human klotho areadded into the wells of a microplate coated with a high affine monoclonal antihumanklotho antibody. During the first incubation step, klotho is bound by theimmobilised antibody. Then the peroxidase-labelled conjugate is added into eachmicrotiter well and a sandwich of capture antibody - human klotho - peroxidaseconjugateis formed. Tetramethylbenzidine (TMB) is used as peroxidase substrate.Finally, an acidic stop solution is added to terminate the reaction. The colour changesfrom blue to yellow. The intensity of the yellow colour is directly proportional to theconcentration of klotho. A dose response curve of the absorbance unit (optical density,OD at 450 nm) vs. concentration is generated using the values obtained fromthe standard. Klotho present in the patient samples is determined directly from thiscurve. |
