| Description | To analyse the G1691A polymorphism, the variable region of the Factor V gene isamplified using real time PCR (e.g. the LightCycler® system). Genomic DNA serves as atemplate for this amplification. Using this amplicon a melting curve analysis is performedthat incorporates specific FRET-hybridisation probes. This melting curve analysisfacilitates the distinct identification of the 1691G or 1691A genotype. For the real time PCRreaction a primer is used that flanks the variable region in the prothrombin gene andproduces a fragment. In addition two probes (fluorescent labeled sequence specificoligonucleotides; anchor and reporter probe) are incorporated into the PCR reaction mix.Both bind the target DNA in close proximity in a region between the primer pair. Upon thisbinding a fluorescent signal is generated that is detected by the optical unit of the real timePCR instrument (e.g. the LightCycler®). For the genotyping analysis a melting curve isrecorded after the amplification reaction. This is achieved by constantly increasing thetemperature. The reporter probe contained in this kit is a perfect match for the wildtypeallele and hence requires a higher temperature to be melted from wildtype DNA than frommutated DNA, as here a mismatch occurs. The sequence of the reporter probe matchesthe non-mutated DNA (1691G) perfectly. The hybrids of probe and target DNA will melt atlower temperatures if the DNA contains the 1691A variant. In case of a heterozygoussample (only one of the two alleles carries the 1691A mutation), two peaks will begenerated, one at the lower temperature (melting point of the mutation) and one at thehigher temperature (melting point of the perfectly matched wildtype). |
