| Description | MutaPLATE® MTHFR (TM) real time PCR Kit contains specific primers and additionalmaterial for the detection of the C677T and A1298C polymorphisms. The variable regionsof the flavin adenine dinucleotide (FAD) binding site of the methylenetetrahydrofolatereductase gene (MTHFR) is amplified by PCR using genomic DNA template astemplate. The specific primers used in the kit flank the variable area and generate anamplificates of 180 bp and 109 bp using TaqMan technology.The standard PCR contains additionally two sequence specific oligonucleotidesmarked with fluorescence dye (TaqMan probes). Both probes bind at the respectiveamplificated target-DNA which includes the single nucleotide polymorphism (SNP). Due tothis, a fluorescence signal is generated and detected by the optical unit of the used realtime PCR instrument. The TaqMan probe for the C- and A-allele (wildtype) is marked withFAM (510 - 530 nm, green) and the TaqMan probe for the T- and C -allele (mutation) ismarked with YAK (555 - 560 nm, yellow). The following three discriminations for each of the polymorphisms are possible:C677T1. Homozygous C/C:Increase of the fluorescent signal from the FAM labeled TaqMan probe, noincrease of the fluorescent signal from the YAK labeled TaqMan probe.2. Heterozygous C/T:Increase of the fluorescent signal from the FAM labeled TaqMan probe andincrease of the fluorescent signal from the YAK labeled TaqMan probe.3. Homozygous T/T:No increase of the fluorescent signal from the FAM labeled TaqMan probe,increase of the fluorescent signal from the YAK labeled TaqMan probe.A1298C1. Homozygous A/A:Increase of the fluorescent signal from the FAM labeled TaqMan probe, noincrease of the fluorescent signal from the YAK labeled TaqMan probe.2. Heterozygous A/C:Increase of the fluorescent signal from the FAM labeled TaqMan probe andincrease of the fluorescent signal from the YAK labeled TaqMan probe.3.
