DescriptionThis antibody reacts with a 25 kDa molecular weight. The antigen is expressed on eosinophils, granulocytes, monocytes and numerous stromal cells. This CD9 antigen belongs to the TM4 superfamily of cell surface proteins with the N- and C-terminals cytoplasmically located. This antibody reacts immunohistochemically on frozen sections of platelets, monocytes and pre-B cells with differential staining on lymphoid and epithelial tissues. CLINICAL RELEVANCE - CD9 mAbs inhibit infection by FIV and CDV (canine distemper virus) but CD9 does not directly bind the virus, suggesting a role as a viral co-receptor, as for diphtheria toxin. - CD9 mAbs have been used for immunophenotyping leukemias and bone marrow purging in autologous bone marrow transplantation - The expression of CD9 is inversely correlated with metastasis in melanoma and breast cancer - CD9 expression is inversely correlated with actuarial survival in lung cancer Higashiyama et al. Cancer Research 55: 6040-6044 1995 - Transfection of CD9 in murine melanoma cells has been shown to reduce metastasis PRINCIPLES OF THE TEST. Immunostep CD9: APC monoclonal antibodies bind to the surface of cells that express the CD9 antigen. To identify these cells, peripheral blood leucocytes are incubated with the antibodies and red blood cells are lysed before washing to remove unbound antibodies. An appropriate fixative solution is added to lysed, washed cells before the stained and fixed cells are analysed by flow cytometry with an argon ion laser at 488nm. REAGENT COMPOSITION. Tetraspanins are cell surface glycoproteins with four transmembrane domains that form multimeric complexes with other cell surface proteins. The encoded protein functions in many cellular processes including differentiation, adhesion, and signal transduction, and expression of this gene plays a critical role in the suppression of cancer cell motility and metastasis.
