The assay uses a caspase-1-specific fluorogenic substrate, Ac-YVAD-AFC. Caspase-1 cleaves this substrate generating free AFC, which can be easily quantified using a fluorescence plate reader at excitation and emission wavelengths of 400 and 505 nm, respectively. The potent and reversible caspase-1 inhibitor Ac-YVAD-CHO is included as a positive control. |
